- [Highly expressed in testis, spleen, colon, and peripheral leukocytes by human CHEK2 Gene (CDS1 Ser/Thr Kinase Family), 543-aa 61-kDa nuclear Serine/Threonine-Protein Kinase CHK2 is a cell cycle checkpoint regulator containing a protein interaction FHA domain for activation in response to DNA damage. Rapidly phosphorylated and activated in response to DNA damage and replication block stress signal transduction, CHEK2 phosphorylates and inhibits CDC25C phosphatase; stabilizes p53 protein leading to G1 cell cycle arrest and preventing mitosis; and phosphorylates BRCA1 allowing survival after DNA damage. CHEK2 defects are associated with Li-Fraumeni syndrome and predisposition to prostate cancer, osteosarcoma, breast cancer, and brain tumors.
