[Encoded by human C5 Gene, Complement Component 5 is a precursor protein containing 1 anaphylatoxin-like domain and 1 NTR domain and similar to C3, C4, and Alpha-2-Macroglobulin. By removal of 4 basic residues, C5 is processed to disulfide linked alpha and beta chains. C5 convertase cleaves the alpha chain, releasing C5a anaphylatoxin and generating C5b. C5 convertase activation of C5 initiates assembly of C5-C9 late complement components into a membrane attack complex; C5b-C6 complex is the assembly foundation of the lytic complex. C5a stimulates chemokinesis and chemotaxis of polymorphonuclear leukocytes to inflammation sites. C5 anaphylatoxin mediates local inflammatory processes; smooth muscle contraction, increased vascular permeability, and histamine release from mast cells and basophilic leukocytes.
