Liposomal NDDP

[A synthetic liposomal formulation of bis-neodecanoate diaminocyclohexane platinum (NDDP) with potential antineoplastic activity. Liposomal NDDP is a third-generation antineoplastic platinum complex, an analogue of cisplatin. After displacement of the 2 long-chain aliphatic leaving groups (neodecanoic acid), platinum diaminocyclohexane (DACH) complexes become highly reactive and able to alkylate macromolecules, forming both inter- and intra-strand DNA crosslinkings, thereby inhibiting DNA synthesis and resulting in cytotoxicity. The DACH moiety of this agent appears to inhibit DNA repair mechanism, which overcome the limits observed in other platinum-based therapies. In addition, the liposomal encapsulation improves the bioavailability of NDDP, increases delivery to tumor tissues and reduces its toxicity profile. ( NCI )]
