[Ataxia Telangiectasia and Rad3 Related Protein (ATR), encoded by the ATR gene, is a member of the PIK (phosphatidylinositol kinase)-related kinase family that are high molecular weight kinases involved in cell cycle progression, DNA recombination, and the detection of DNA damage. ATR is essential for the viability of human somatic cells. ATR and ATRIP (ATR-interacting protein) are mutually dependent partners in cell cycle checkpoint signaling pathways - ATR phosphorylates ATRIP that regulates ATR expression and is an essential component of the DNA damage checkpoint pathway. ATR phosphorylates checkpoint kinase CHK1, checkpoint proteins RAD17, and RAD9, as well as tumor suppressor protein BRCA1.
