[MPL encodes the Thrombopoietin Receptor, a member of the cytokine receptor superfamily. The mechanism by which TPO activates the TPOR appears similar to that of other hematopoietic cytokines that bind and induce receptor homodimerization. TPO regulates the production of multipotent hematopoietic progenitor cells and platelets. Lymphoid progenitors express IL7R and not MPL, whereas myeloid progenitors express MPL and not IL7R. The receptor for thrombopoietin may also represent a regulatory molecule specific for TPOR-dependent immune responses. TPOR contains two fibronectin type III-like domains and is a type I membrane protein. Two isoforms, MPL-K and MPL-P, are produced by alternative splicing and are co-expressed.
